Unfortunately there is no subway or a train going straight from the airport to the center of Prague. So the MHD buses and the Airport express bus line are the only options going from the airport to the center of town. Use the 119 or 100 bus line for the fastest transport to town or to the nearest subway station. If you need to get to the train station, use the Airport express. Also if you arrive to Prague after 11pm, there are no MHD buses going anymore. You would have to use the tram or a night bus line, but it can take you almost 2 hours to get where you need to. If you use the 119 bus line, you should go to the last station, which is Nadrazi Veleslavin, where you can enter the subway line A and go to the stations Malostranska, Staromestska, Mustek or Muzeum. They all are in the city center. If you use the 100 bus line, from the airport you will get to Zlicin where you can use the subway line B. It takes about 30 minutes and you should get out at Mustek or Florenc station for the city center.

You can also use all these options going the other way to the Prague airport. So the best option from the city center is probably to use the Subway line A to Nadrazi Veleslavin station and then the 119 bus line to the airport terminals.
You can buy the tickets right at the arrival halls at both terminals and they are open from 7am to 10pm. You can also find a ticket machine at every bus station at the airport or you can buy tickets directly from the bus driver but those are more expensive. Also make sure that you mark the ticket at the bus station or in the bus. You can use te ticket markers that are placed near the handles right next to the bus doors. In subway stations the markers are placed by the entrance to the station.
